摘要
目的:探讨齿状回(dentate gyrus,DG)一氧化氮合酶(NOS)与反复游泳应激的关系,观察了反复游泳应激对大鼠齿状回神经元型一氧化氮合酶(nNOS)表达的影响。方法:经4周大负荷游泳训练制成大鼠应激模型,于安静状态下,用ABC免疫组织化学方法显示齿状回nNOS阳性神经元,用图像处理半定量方法对nNOS阳性神经元的数量、面积和灰度进行测量和分析。结果:应激组大鼠齿状回nNOS阳性神经元的数量、面积和灰度均明显大于对照组(P<0.01,P<0.05)。结论:反复游泳应激可使大鼠齿状回神经元中的nNOS上调,齿状回nNOS神经元参与了中枢运动性疲劳的形成,其机理可能与齿状回对应激反应的心理行为调节以及NO的神经毒性有关。
Objective: To explore the relationship between repeated swim stress and nitric oxide syntyase (NOS) in dentate gyrus, it is observed that the effects of repeated swim stress on expression of neuronal nitric oxide syntyase (nNOS) in rat dentate gyrus. Methods: After long term (4 weeks) swimming exercise with high intensity, the rats stress model is made. In a quiet state, nNOS immunpoositive neurons in dentate gyrus is shown by using ABC immunohistochemistry integrating with image pattern semiquantitative analysis (neuron number, area and gray degree). Resuits: A significant increase of nNOS immunopositive neurons in dentate gyrus in the number, area and gray degree in stress group (P 〈 0.01, p 〈 0.05). Conclusion : Repeated swim stress induces up - regulation of nNOS in rat dentate gyrus, which is involved in forming exercise - induced central fatigue, due to the psychological and behavioral regulation of dentate gyrus on stress as well as NO. neurotoxicitv.
