应用体外转录法和荧光素制备RNA探针的技术

The Technique of Preparing RNA Probe by in vitro Transcription and with Fluorescein

为建立一种快速制备RNA探针的方法,以研究不同的RNA与RNA之间的相互作用,介绍一种用体外转录法制备RNA,然后用聚丙烯酰胺凝胶电泳法(PAGE)纯化得到纯化的RNA,最后用荧光素标记得到RNA探针的新方法,用该方法制备RNA探针简便、高效、安全,尤其适合小剂量RNA探针的制备。

With the rapid development of modern biological science, our research has reached the molecular level already and at the same time more and more people focus on RNA research. However the concentration of RNA is so small that it is difficult to detect and to observe RNA and their interaction. The technique we report here uses one fluorescent material to label RNA, then we can detect and observe RNA and their interaction easily. Firstly we make RNA in vitro transcription and purify it by PAGE,finally label RNA with fluorescein. Labelled RNA must be purified by PAGE or HPLC because the mixture after reaction contains fluorescein RNA probe and unlabelled RNA,that is the RNA probe we want to get. As far as the technique is concerned, it is more convenient, efficient and safe. It is very useful for making a small dose of RNA probe especially.
