摘要
目的使用基因芯片对BEL-7402细胞系和正常肝组织的基因表达谱作分析。方法提取BEL-7402细胞及正常肝组织RNA逆转录用dUTP-cy5、dUTP-cy3分别标记cDNA探针,用基因芯片检测与肿瘤相关的866个基因的表达。结果BEL-7402细胞系与正常肝组织比对发现31个基因有表达差异;其中上调24个,下调7个。这些差异表达的基因多与细胞周期调控、受体表达及免疫相关。结论从实验中发现的表达差异的基因有利于干预癌细胞周期调控和信号通道的各个环节,为肿瘤药物的研发和治疗提供基本信息。
[Objective] To investigate the molecular characters of BEL-7402 cell line. [Methods] Compared gene expression changes of BEL-7402 cell line with normal liver tussue by cDNAmicroarray among 866 cancer related genes. [Results] There were 31 genes had differential expression in BEL-7402 cell llne. 24 genes were up- regulated while 7 genes were down-regulated. In those genes, many differentially expressed genes were involved in cell division,membrane receptor and immunocompeten. [Conclusions] Abnormal gene expression contribute to mahgnant cell divison.
