白介素-10对脊髓损伤后炎症影响的研究 被引量:6

摘要 目的研究IL-10对脊髓损伤(SCI)后NF-κB表达和单核/巨噬细胞浸润的作用及意义。方法制作大鼠SCI模型,伤后半小时分别给予IL-10(实验组)和生理盐水(对照组)腹腔注射,常规方法提取脊髓组织核蛋白完成核蛋白定量,Trans AMTM NF-κB P65试剂盒检测NF-κB P65表达。结果两组脊髓组织中均有NF-κB P65表达,两组表达水平具有显著差异(P<0.05,<0.01);两组单核/巨噬细胞数目和密度具有显著差异(P<0.01)。结论IL-10对脊髓损伤后炎症介质核因子NF-κB表达和局部单核/巨噬细胞浸润有抑制作用,两种机制共同作用有效抑制了SCI早期炎症的发生。 Objective To investigate the implication and effect of IL-10 for the expression of NF-κB and the infiltration of monocyte/macrophage after spinal cord injury. Methods A model of spinal cord injury was completed. Half an hour later, each group had been administered peritoneally IL-10 in experimental group, PS in control group, respectively. The neueleopretein of myeloid tissue was extracted in routine way and then the neueleopretein was quantitated. The expression level of NF-κB P65 in myeloid tissue was analyzed with a kit ( Trans AMTM NF-κB P65 ). Results The expression of NF-κB P65 existed in myeloid tissue of the experimental group and the control group. There was obviously different in the expression levels of NF-κB P65 in two groups ( P 〈0.05 or P 〈0.01 ). The cell population and cell density of Monocyte/ Macrephage remained remarkably different in both groups ( P 〈 0.01 ). Conclusion IL-10 has depressant effect for expression of NF-κB and monocyte/macrephage local infiltration in spinal injury, two mechanisms contribute to the active suppression in earlier period of inflammation occurrence in spinal cord injury.
