氧化锌对人呼吸道上皮细胞IL-8表达作用

摘要 目的探讨氧化锌对人呼吸道上皮细胞白细胞介素-8(IL-8)表达的影响及其分子生物学机制。方法以人呼吸道上皮细胞作为体外模型,应用PCR与ELISA方法测定氧化锌诱导IL-8mRNA的表达及蛋门的释放,以及表皮生长因子(EGF)受体的磷酸化。结果氧化锌可诱导呼吸道上皮细胞过量表达IL-8mRNA。在同一试验条件下,氧化锌可诱发EGF受体磷酸化。结论EGF受体及细胞吞噬作用在氧化锌诱导呼吸道上皮细胞IL-8表达过程中起重要作用。 Objective To explore the effect of zinc oxide(ZnO)-induced IL-8 expression in human airway epithelial cells and underlying mechanisms.Methods Human airway epithelial cells(BEAS-2B)were employed as airway model in vitro.Expression of IL-8 mRNA and protein was determined by PCR and ELISA assay.Results Exposure to ZnO of airway epithelial cells resulted in overexpression of IL-8 mRNA and subsequent overproduction of IL-8 prote in.ZnO stimulation was shown to induce phosphorylation of EGF receptor,indicative of EGF recep tor activation.Conclusion Zinc oxide exposure activates EGF receptor.Both EGF receptor and phagocytosis are involved in the regulation of ZnO-induced IL-8 expression.
