砷和香烟烟气溶液对基因Bcl/2和Bax表达影响 被引量:3

摘要 目的研究亚砷酸钠和香烟烟气溶液联合作用对凋亡相关基因Bcl-2和Bax表达的影响,并探讨它们对这2种基因表达的影响是否存在交互作用。方法按照2×2析因设计将大鼠淋巴细胞分为4组:亚砷酸钠单独作用组、香烟烟气溶液单独作用组、两者联合作用组和对照组。利用免疫印迹法检测细胞的Bcl-2和Bax表达,利用磷脂酰丝氨酸外翻分析法检测各组细胞凋亡,同时采用2×2析因设计的方差分析研究两者的交互作用。结果亚砷酸钠和香烟烟气溶液单独作用或者联合作用都能够使细胞Bcl-2蛋白表达减少、Bax蛋白表达增加、凋亡细胞数增加。析因设计的方差分析结果表明,2种毒物对Bcl-2蛋白表达、Bax蛋白表达、凋亡细胞数均存在交互作用,交互作用方式为协同作用。结论亚砷酸钠和香烟烟气溶液对大鼠淋巴细胞的Bcl-2蛋白表达、Bax蛋白表达的影响存在交互作用,交互作用方式为协同作用。 Objective To investigate the effect of sodium arsenite combined with cigarette smoke solution on expression of Bcl-2 and Bax in rat lymphocytes and study their interaction.Methods According to 2×2 factorial design,rat lymphocytes were divided into 4 groups:sodium arsenite treated group,cigarette smoke solution treated group,sodium arsenite and cigarette smoke solution treated group and control group.After the exposure,protein expression of Bcl-2 and Bax in rat lymphocytes was detected by western blotting;Apoptosis of rat lymphocytes was detected by flow cytometry.ANOVA of 2×2 factorial design was used to evaluate the interaction.Results When sodium arsenite treatment and cigarette smoke solution treatment were used together or separately,protein expression of Bcl-2 decreased;Protein expression of Bax and the number of apoptotic cells increased.ANOVA of the factorial design showed that sdium arsenite and cigarette smoke solution had interactions on protein expression of Bcl-2,Bax and the number of apoptotic cells.The interactions were synergistic.Conclusion Sodium arsenite and cigarette smoke solution have interactions on protein expression of Bcl-2 and Bax.The interactions are synergistic.
