摘要
本工作在原代培养的小鼠肝细胞上观察了一氧化氮(NO)、cGMP和CAMP的变化与神经降压素肝细胞保护作用之间的关系。结果如下:向培养液中加入酪氨酚(20mmol/L)12h后,GOT和GPT漏出明显增加;在加醋氢酚之前给予神经降压素(10-7mol/L)则使转氨酶漏出明显减轻。给予NO合成酶阻断剂L-NAME可完全阻断神经降压素的保护作用。神经降压素使肝细胞内cGMP含量显著增加,但对cAMP含量无明显影响。结果表明,神经降压素的肝细胞保护作用是由NO介导的,且与cGMP有一定关系。
In the present work, the relation of cytoprotection of neurotensin to nitric oxide(NO), cGMP and cAMP was investigated in primary cultured mouse hepatocytes. Theresults are as follows: After administration of acetaminophen (20 mmol/L) to the medium, the leakage of GOT and GPT increased significantly. Pretreatment with neurotensin (10-7 mol/L) before acetaminophen reduced the leakage of GOT and GPT.No synthase inhibitor, L-NAME, completely blocked the cytoprotective effect of neurotensin. Neurotensin could enhance the intracellular cGMP content, but had no effecton cAMP content. These results indicate that the protective effect of neurotensin on hepatocytes is mediated by NO probably by enhancing intracellular cGMP content.
