广元地区宫颈癌组织人乳头瘤病毒感染及其基因变异调查 被引量:5

摘要 [目的]了解广元地区妇女宫颈癌组织人乳头瘤病毒(HPV)感染状况和优势HPV亚型,分析HPV16E6基因的变异及地域株分布。[方法]对宫颈癌的患者,采用HPV普适引物GP5+/GP6+和高危HPVE6基因特异引物相结合的方法,对其肿瘤组织中所提取DNA进行HPVPCR扩增,扩增片断经DNA测序确认其HPV感染率及亚型。[结果]HPV总体感染率为97.2%;HPV16为主要型别,占78.5%,其中HPV16单一感染23例,与其他HPV亚型共感染32例;HPV58感染率为第2,占27.1%;其余依次分别为HPV59(21.4%,);HPV52(20.0%,),HPV18(11.4%,);HPV33,HPV45和HPV6各1例(1.4%)。共有38例肿瘤组织中检测到不同HPV型别的共感染,占总感染的54.3%。感染的HPV16中,东亚株占34.5%,欧洲株占32.7%)。[结论]该地区宫颈癌组织中HPV感染以16型为优势感染亚型,HPV58,59和52为亚优势感染型别,超过半数患者为混合感染。HPV16感染中以HPV16东亚和欧洲流行株为主。 [Objective] To investigate the infection status and predominant genotypes of human papillomavirus (HPV) of the patients with cervical cancer and to analyze the mutations and the geographical distribution of HPV16 E6 gene in Guangyuan area. [ Methods] The DNA samples from 72 cases of cervical cancer dssues were detected for HPV infection by using the combinative method of general primer-med/ated GP5+/GP6+ PCR and E6 specific primer-mediated PCR. [Resolts] The total infection rate of HPV was 97.2% ; HPV16 was the most common type with infection rate of 78.5%. Among the cases infected with HPV16, 23 cases were infected with HPV 16 alone, and 32 eases were infected with multiple subtypes of HPV. The infection rate of HPV58 ranked the second accounting for 27.1%. The rest were HPV59 (21.4%). HPV52 (20.0%), HPV18 (11.4%), HPV33 (1.4%) and HPV45 (1.4%). 38 cases were infected by multiple HPV types (54.3%). Among the HPV16 variants infected, East Asia type was 34.5% and European type was 32.7%. [Conclusion] HPV16 is the most prevalent HPV type in cervical cancer tissues of the patients in Guangyuan area, followed by HPV58, 59 and 52. Over half of patients' infections are combined infections. East Asia and European type of HPV16 are popular variants in HPV16 infection.
