麝香酮对连二亚硫酸钠造成PC12细胞缺氧损伤的保护作用 被引量:19

摘要 目的:研究麝香酮对PC12细胞缺氧损伤的影响。方法:在离体培养的PC12细胞用连二亚硫酸钠造成缺氧损伤模型,通过细胞形态学观察、MTT微量比色、培养介质LDH活力测定,研究了麝香酮对该模型的保护作用。结果:5×10-7~510-51mol/L范围内,麝香酮浓度依赖性地降低连二亚硫酸钠造成缺氧损伤模型所致培养介质内LDH的释放,其IC50为43.91μmol/L。在10-7~5×10-4mol/L范围内,麝香酮浓度依赖性地增加连二亚硫酸钠造成缺氧损伤模型培养介质MTT比色值。其IC50为28.31μmol/L。结论:麝香酮对连二亚硫酸钠造成PC12细胞缺氧损伤具有保护作用。 Objective:To investigate the effects of muskiness ketone on anoxia injury in cultured PC12 cells. Methods: Cultured PC12 cells were treated with 1 mmol/L Na2 S2O4 in combination with glucose deprivation. The protective effects of muskiness ketone on the model were evaluated hy lactate dehydrogenate (LDH) efflux assay and colormetric MTT assay. Results: Muskiness ketone, within the range of 10^-7-5 ×10^-5 mol/L significantly antagonized LDH efflux induced by the model and increase the optical density at 570nm tested by colorimetric MTT assay in concentration-dependent manner. Conclusion: It was shown that muskiness ketone markedly protected PC12 ceLLs from anoxia injury.
