摘要 本实验旨在研究高压脉冲电场(PEF)对多聚半乳糖醛酸酶(PG)活性及构象的影响。PG的活性随电场强度和脉冲个数的增加而降低。荧光光谱分析表明:PEF处理后PG蛋白的荧光发生不同程度的荧光淬灭,在高电场条件下将发生荧光峰的位移。另外,PG酶活性下降和荧光淬灭趋势一致,但呈非线性关系,说明酶构象改变与酶活性变化间存在密切的关系。 Effect of high pulsed electric field on the activity and conformation of polygalacturonase(PG) was studied in this paper.The activity of PG was decreased with increasing electric field intensity and pulsed number.Spectrofluorimetry showed that the fluorescence of PG quenched in different degree by pulsed electric field.The place of peak fluorescence intensity changed under high electric field intensity.The trend of activity decreased and was similar to fluorescence quenching trend.But the detail relationship was nonlinearity.All these indicated that changes of PG conformation had a close relation with the inactivation.
