摘要
研究了高效液相色谱法同时测定啤酒酵母RNA降解产物4种5'-核苷酸的适宜条件。应用HypersilODS2色谱柱,在260 nm检测波长下,以0.08 mol/L KH2PO4缓冲溶液(pH2.5)作流动相,10 min内可完成CMP、UMP、AMP、GMP含量的同时测定,最小检测限分别为0.2 mg/L、0.2 mg/L、0.1 mg/L和0.3 mg/L,平均回收率分别为99.21%、100.39%、100.07%和99.88%,相对标准偏差(RSD)分别为2.27%、2.83%、2.05%和1.85%。
The optimum determination conditions of four kinds of 5′-nucleotides (beer yeast RNA degradation product) by HPLC simultaneously were studied. The measurement of CMP, UMP, AMP and GMP content could be finished within 10 min as ODS2 column was used as column, 0.08 mol/L KH2PO4 (pH=2.5) was used as the mobile phase, and the detection wave-length was set at 260 nm. The results showed that the mini-mum detection limit were 0.2 mg/L, 0.2 mg/L, 0.1 mg/L and 0.3 mg/L respectively and the average recovery were 99.21%, 100.39 %, 100.07 % and 99.88 % respectively and RSD were 2.27 %, 2.83 %, 2.05 % and 1.85% respectively.
