摘要
目的建立白芍药材中白芍总苷的含量测定方法。方法以碱水解法将白芍总苷水解为苯甲酸,采用高效液相色谱法,以十八烷基硅烷键合硅胶为固定相,乙腈-0.1%磷酸(25∶75)为流动相,检测波长230nm,流速1.0mL/min,测定苯甲酸含量,从而计算出白芍总苷含量。结果方法重现性和稳定性均良好,苯甲酸进样量线性范围为20.016~500.4ng,回归方程为Y=5.29613X-1.06420,r=1.0000(n=5),平均回收率为98.58%,RSD为0.95%(n=9)。结论该方法适用于白芍药材中白芍总苷的含量测定。
Objective To establish a HPLC method for the determination of total glucosides of paeony in Radix Paeoniae Alba. Methods Using the method of alkaline hydrolysis,the total glueosides of paeony were hydrolyzed into benzoic acid. Using a HPLC method with 18 alkyl silane Bonded Silica as the stationary phase,the mobile phase was a mixture of acetonitrile- 0. 1% phosphoric acid (25:75), the detection wavelength was 230 nm, the flow rate was 1.0 mL/min. The content of total glucosides of paeony could be determinated by benzoic acid. Results The repeatability and stability were fine. The content of benzoic acid showed a good linear relationship in the range of 20.016-500.4ng (r=l. OOOO, n=5).The average recovery rate was 98.58%,RSD was 0.95% (n=9).Condusion The methods can be used to determine the total glucosides of paeony in Radix Paeoniae Alba.
