蝎毒素-A中枢镇痛作用的研究

THE STUDY ON THE EFFECT OF MICROINJECTION OF SCORPION VENOM A INTO PERIAQUEDUCTAL GRAY IN ANALGESIC

目的研究蝎毒及其提取物的中枢镇痛作用。方法向大鼠中脑导水管周围灰质（Ｐｅｒｉ－ａｑｕｃｔａｌＧｒａｙ，ＰＡＧ）内微量注射蝎毒素－Ａ（ＳｃｏｒｐｉｏｎＶｅｎｏｍ－Ａ，ＳＶ－Ａ），以热辐射甩尾法为指标，观察其中枢镇痛作用效果。结果向大鼠ＰＡＧ内匀速注射０．０１２５％～０．０５％ＳＶ－Ａ０．５μｌ（相当于０．２５～１μｇ·ｋｇ－１）即出现痛阈升高，２０ｍｉｎ达峰值（痛阈最大提高１５０％以上），与对照组比较差异有显著性（Ｐ＜０．０１）。本项研究工作证实，欲使大鼠痛阈提高１５０％，ＰＡＧ内注射吗啡的用量约为１０μｇ·ｋｇ－１。结论ＳＶ－Ａ有很强的中枢镇痛作用，以达相同镇痛效能而论，ＳＶ－Ａ的中枢镇痛作用强于吗啡１０倍。本品属中枢性强镇痛剂。

Objective To study central analgesic effect of Scorpion Venom and its extrat.Method A thorough center analgesic effect is reported in this paper through periaqueductal gray(PAG) ingested Scorpion Venom A(SV A) evaluated by flexion reflex radiated by the heat in rat.Result The pain threshold increases at the peak after 20 minutes of injecting 0.25～1μg·kg 1 SV A(increase by 150 percent).This has a significant diffference from the control group (P<0.01).It was reported that ingecting 10μg·kg -1 Morphine through PAG will obtain the same potency.That is to say,the potency of SV A is 10 times greater than that of Morphine.Conclusion The above show that SV A is a potent cenrtal analgesic.
