高效液相色谱法测定蒙药草乌花中药效组分的含量 被引量:4

摘要 目的:建立以高效液相色谱法同时测定蒙药草乌花中乌头碱、新乌头碱、次乌头碱含量的方法。方法:Extend-C18(250mm×4.6mm,5μm),流动相为甲醇-0.3mol/L三乙胺(65∶35),流速为0.8mL/min,检测波长为235nm,柱温为35℃。结果:乌头碱、新乌头碱、次乌头碱分别在1~5μg,0.35~1.75μg,0.5~2.5μg范围内呈良好的线性关系。结论:本方法简便、准确、分离效果好、线性范围宽、灵敏度高,可用于本品的质量控制。 Objective: To establish an HPLC method for simultaneous content determination of aconitine, mesaconitine, and hypaeonitine in Flos Aconiti kusnezoffii. Methods: Extend -C18 (250mm ×4. 6mm ,5 μm) was used as chromatographic column, methanol - 0.3 mol/L triethylamine ( 65 : 35 ) as mobile phase. The detection wavelength was set at 235 nm, the flow rate at 0. 8mL/min and the column temperature at 35℃. Results :The sample sizes of aconitine, mesaconitine, hypaconitine had good linear relationship with their peak areas in the range of 1 - 5μg, 0.35 - 1.75μg, and 0.5 - 2.5 μg, respectively. Conclusion :The established method is convenient and accurate with satisfactory separation effect,wide linear range, and high sensitivity ,and can be available for the quality control of Aconitum kusnezoffii Reichb.
