摘要 目的:探讨中药对过氧化氢(H2O2)诱导人脐静脉内皮细胞株(ECV-304)损伤的保护作用及其可能机制。方法:对体外培养的ECV-304,采用电镜及流式细胞术等方法,观察过氧化氢的损伤方式及中药对其的影响。结果:过氧化氢组(30.735±11.38)与对照组(9.24±1.828)比较,细胞凋亡明显增加,差异显著(P<0.01)。中药干预组(20.48.±7.12)与过氧化氢组(30.735±11.38)比较,明显下降,差异显著(P<0.01)。与对照组(9.24±1.828)比较,有差异,下降不明显。结论:过氧化氢刺激后(200mmol/L),内皮细胞功能,结构发生明显变化。损伤形式主要表现为活性氧的应激,对死亡受体通路的活化而导致细胞凋亡速度加快。中药可抑制活性氧对血管内皮细胞的损伤,对血管内皮细胞具有一定的保护作用。 Objective :Approach on the protection and possible mechanism of traditional Chinese medicine on the injured human umbiblical vein endothelial cell line (ECV -304) induced by hydrogen peroxide. Methods :Electron microscope and flow cytometry etc. are used to observe the injured pattern of hydrogen peroxide and the effect of traditional Chinese medicine on the ECV - 304 cultured in vitro. Results : Hydrogen peroxide group comparing with the control group, apoptosis obviously increase, significant difference ( P 〈 0.01 ). Traditional Chinese medicine intervention group comparing with the hydrogen dioxide group, apoptosis obviously decrease, significant difference ( P 〈 0.01 ). Traditional Chinese medicine intervention group comparing with the control group, the difference isnt significant. Conclusions :Hydrogen peroxide (200mmol/L) can result in the marked changes of the function and the construction of endothelial cell. The injured patterns mainly include the stress of the active oxygen and the activation of the death receptor channel, which can result in the acceleration of apoptosis. Traditional Chinese medicine can inhabit the active oxygen injuring the vascular endothelial cell and has some protection on the vascular endothelial cell.
