摘要
以授粉后黄瓜幼果组织的总RNA为模板,利用简并引物,采用RT-PCR方法扩增出1条长度为480 bp的cD-NA片段。序列分析结果表明,该cDNA片段与来源于黄瓜根组织的CsEXP5片段序列(AF319471)同属一个基因,序列拼接后得到526 bp的cDNA序列。预测的CsEXP5蛋白具有一系列保守的Trp残基和HFD,KNFRV保守域。该蛋白属于α-扩张蛋白,与CsEXP10蛋白的同源性高达90%。该基因首次从黄瓜幼果中得以克隆,可能与黄瓜果实膨大生长有一定关系。
A new cDNA fragment of 480 bp was amplified by RT-PCR from total RNA of cucumber young fruits after pollination. Sequence analysis showed that the cDNA fragment was the same gene to CsEXP5 (AF319471)from cucumber roots, and a 526 bp cDNA fragment was obtained by splicing. The predicted CsEXP5 protein contains a series of conserved Try and HFD, KNFRV conserved domains. CsEXP5 was α-expansin and shares 90% identity with CsEXP10. CsEXP5 might be correlated with the expansive growth of cucumber fruit.
