摘要 [目的]探讨雌二醇对宫颈癌细胞株Hela和Siha基质金属蛋白酶(MMP)2和9mRNA转录水平的影响。[方法]采用RT-PCR检测雌二醇(1×10-8mol/L)干预72h后两株细胞MMPs表达的变化。[结果]Hela细胞株表达MMP-9,但不表达MMP-2,而Siha细胞相反。雌二醇干预后,Hela细胞MMP-9mRNA表达明显高于对照组(P=0.004),Siha细胞MMP-2mRNA的表达与对照组的无显著性差异(P=0.409)。[结论]MMPs在宫颈鳞癌和腺癌中可能表达不同的亚型,1×10-8mol/L的雌二醇能够促进Hela细胞MMP-9mRNA的转录。 [Purpose] To investigate the effects of 17β-estradiol (E2) on the expression of matrix metalloproteinase-2 and 9 (MMP-2, MMP-9) mRNA in cervical cancer cell lines(Hela and Siha). [Methods] After the cell lines incubated with E2 at 1×10^-8 mol/L for 72hrs, the expression of MMPs mRNA were detected by reverse transcription-polymerase chain reaction (RT-PCR). [Results] MMP-9, but not MMP-2, was expressed in Hela cells. By contrast, only MMP-2 was expressed in Siha cells. After exposure to E2, the level of mRNA expression of MMP-9 in Hela cells increased significantly compared with the control group (P=0.004). However, the expression of MMP-2 mRNA of siha cells in the 2 groups was not significantly different (P=0.409). [Conclusion] MMPs might express different subtypes in squamous carcinoma and adenocarcinoma of cervix. Exposure to E2 at 1×10^-8mol/L may promote MMP-9 mRNA transcription.
