摘要
目的采用AdEasy系统构建的带有标记基因RFP的重组腺病毒Ad-Cyr61感染人肝癌细胞株HepG2,并观察外源性Cyr61对肿瘤细胞生长和迁移的影响。方法Ad-Cyr61与Ad-RFP分别感染人肝癌细胞株HepG2,荧光倒置显微镜观察荧光表达,通过RT-PCR、免疫组化实验分别检测Ad-Cyr61的mRNA和蛋白的表达;通过MTT实验、细胞计数实验、克隆形成实验和划痕愈合实验,观察Cyr61对人肝癌细胞株HepG2细胞增殖和迁移的影响。结果Ad-Cyr61感染人肝癌细胞株HepG2,与对照组比较细胞数量增多,集落形成增加,细胞划痕逐渐愈合。结论Ad-Cyr61感染人肝癌细胞株HepG2后可促进细胞增殖和迁移,提示Cyr61在肿瘤发生和转移中可能起到重要作用。
Objective To observe the proliferation and migration of human hapetocelluar carcinoma cell line HepG2 after the introduction of exogenous cysteine rich 61 (Cyr61). Methods Adenviral vector, AdEasy with a report gene, red fluorescence protein (RFP) was used to construct recombinant vector Ad-Cyr61. The vectors Ad-Cyr61 and Ad-RFP (control) were transformed into HepG2 cells to observe the protein and mRNA expression of Cyr61 by immunohistochemistry and RT-PCR. MTT, cell counting, plate colony formation and cell wound healing assay were performed to determine the Cyr61 ' s effect on the proliferation and migration of HepG2. Results Ad-Cyr61 transfected HepG2 cells expressed exogenous Cyr61, formed more clones, proliferated more cells and exhibited a greater ability to migrate in cell wound healing than the control cells. Conclusion Exogenous Cyr61 stimulates the proliferation and migration of HCC cells, suggesting that Cyr61 may play a role in tumorigenesis and metastasis.
