Expression of Fas/FasL in Colorectal Carcinoma and Their Associations with Development and Metastasis of Colorectal Carcinoma

摘要 【目的】探讨Fas/FasL在大肠癌组织细胞的表达以及它们与肿瘤的发展及转移的关系。【方法】应用免疫组化(SABC)法对41例(包括各个不同Dukes分期)的大肠癌组织以及正常大肠组织的Fas/FasL进行检测。【结果】大肠癌组织Fas阳性率为48.8%,低于正常大肠黏膜组织的Fas阳性率92.7%(P<0.01)。大肠癌组织FasL阳性率为53.7%,高于正常大肠黏膜组织的FasL阳性率4.9%(P<0.01)。在不同分期的大肠癌组织中,Fas阳性率分别为:A期87.5%,B期66.7%,C期25.0%,D期21.2%。而FasL阳性率分别为:A期25.0%,B期33.3%,C期66.7%,D期88.8%。随着Dukes分期的递升,Fas阳性率呈递减趋势,而FasL阳性率呈递升趋势。【结论】在大肠癌组织中,Fas表达下调,以及FasL表达上调,肿瘤细胞凋亡减少,这有利于肿瘤细胞的增殖。Fas/FasL在大肠癌的发生、发展和转移中起着重要的作用,为大肠癌的基因治疗提供了新思路。 [Objective] To study the expression of Fas/FasL in colorectal carcinoma and their associations with the development and metastasis of colorectal carcinoma. [Method] By using the immunohistochemical methods (SABC), we analyzed 41 cases of colorectal carcinoma, which included different Dukes stage cases, and 20 cases of normal colorectal tissues. We detected the expression rates of Fas/FasL in those cases. [Result] The expression rates of Fas in the colorectal carcinoma was 48.8%, which was significantly lower than the rate of 92.7% in the normal colorectal tissues (P 〈 0.01). The expression rates of FasL in the colorectal carcinoma was 53.7%, which was significantly higher than the rate of 4.9% in the normal colorectal tissues (P 〈 0.01). In the different Duke stages, the rates of Fas (+) were: A=87.5%, B=66.7%, C=25.0%, D=21.2%. The rates of FasL (+) were: A= 25.0%, B=33.3%, C=66.7%, D=88.8%. The expression rates of Fas and FasL were closely related to the Dukes stage. The higher the tumor stage was, the lower expression rates of Fas were, and the higher the expression rates of FasL were. [Conclusion] The increased expression of FasL and the decreased expression of Fas both played important roles in the development and metastasis of colorectal carcinoma. These may serve as new approaches for us to do some genetic therapy in metastasis of colorectal carcinoma.
