目的探讨p27Kip1基因在骨肉瘤MG-63、U2细胞株及骨肉瘤组织中的表达情况及其与骨肉瘤生物学行为的关系。方法培养人骨肉瘤细胞MG-63、U2和人成骨细胞OB;以半定量RT-PCR技术检测3种细胞中p27Kip1基因的mRNA表达情况。以免疫细胞化学染色技术检测MG-63、U2及OB中p27Kip1蛋白的表达情况。以免疫组织化学方法检测骨肉瘤、骨软骨瘤、正常骨3种组织中p27Kip1蛋白的表达情况,并探讨其与临床病理学特征之间的关系。结果p27Kip1基因mRNA的表达在人骨肉瘤细胞MG-63、U2和人成骨细胞OB中没有差别。p27Kip1蛋白的表达在OB中高于MG-63和U2,MG-63和U2间p27Kip1蛋白的表达没有差别。骨肉瘤组织的p27Kip1蛋白表达低于骨软骨瘤组织和正常骨组织,p27Kip1蛋白表达与Enneking外科分期及是否发生转移相关。结论骨肉瘤细胞、组织p27Kip1蛋白表达明显降低;p27Kip1蛋白的表达可作为骨肉瘤预后判断的一个有价值的指标。

Objective To detect the expression of gene p27^Kip1 in osteosarcoma cell line MG-63, cell line U2 and tumor tissue, and to investigate the correlation of the gene expression with the biologic behaviour of ostosarcoma. Methods 1. the expression of the mRNA in gene p27^Kip1 in cultured osteosarcoma cell line MG-63, cell line U2 and osteoblast cell line OB was detected with semi-quantitative RT-PCR technique. 2. The expression of protein p27^KIP1 in these cell lines was detected with immunocytochemical staining. 3. We detected and compared the expression of protein p27^KIP1 in osteosarcoma, ostochondroma and normal bone tissue by immunohistochemical staining, and explored its correlation with the clinical patholigical characters of ostosarcoma. Results 1. There no difference of the expression of mRNA p27^KIP1among the cell lines MG- 63, U2 and OB. The expression of protein p27^KIP1 in cell line OB was higher than that in cell lines MG-63 and U2 with no difference between them. 2. The expression of protein p27^KIP1in osteosarcoma tissue was lower than that in osteochondroma and normal boon tissue. The declined expression had correlation to the Enneking Grading and metastasis. Conclusions The expression of protein p27^KIP1 in osteosarcoma cell lines and tissue decrease significantly. The expression of protein p27^KIP1 can be regarded as an indicator for the prognosis of osteosarcoma.
