摘要
目的:探讨低氧诱导因子1α(HIF-1α)对小鼠宫颈癌皮下移植瘤放射敏感性的影响及其可能的机制。方法:建立宫颈癌皮下移植瘤模型,随机分为对照组、单次γ射线大剂量照射组、小剂量分次照射组、大剂量照射联合高压氧组、小剂量分次照射联合高压氧组,采用不同的方案进行体外放射治疗。免疫组化法测定放射治疗后移植瘤内的HIF-1α、Bcl-2的表达水平,观察各γ射线处理组照射后移植瘤的抑瘤率,并对HIF-1α与Bcl-2的表达及抑瘤率进行相关性分析。结果:HIF-1α的表达与移植瘤抑瘤率呈负相关(r=-0.514,P<0.05);与Bcl-2表达无明显相关关系。结论:抑制HIF-1α的表达可提高肿瘤对放射治疗的敏感性,该效应可能与Bcl-2表达无关。
Objective: To study the effect of of hypoxia - inducible factor - 1 alpha on the radiosensitivity of rat cervical carcinoma and discuss the mechanism. Methods: The transplantation tumor model of cervical carcinoma was established. 77 rats were randomly divided into five groups : control group, the large dose single - γ - ray irradiation group, the small dose fraction irradiation group, the large dose irradiation combined HBO group and the low dose irradiation combined HBO group. Expression of HIF -1α and Bcl -2 were detected by immunohistochemical staining, The tumor inhibitory rate was measured by the change of tumor volume. The correlativity between the expression of HIF -1α and Bcl -2 and radiosensitivity was observed. Results: Expression of HIF -1α and carcinoma growth inhibitory rate was correlated negatively ( r= - 5. 147, P 〈 0. 05 ) . There was no significant correlation between the expression of HIF -1α and Bcl - 2. Conclusion: Inhibition of the HIF -1α expression can effectively increase the radiosensitivity of rat cervical carcinoma , this effect have no relation with the expression of Bcl -2.
