不同程度ICP患者胎盘中NPY的表达及临床意义

Expression and clinical significance of neuropeptide (NPY) in placenta of patients with intrahepatic cholestasis in various clinical stages

目的:探讨妊娠肝内胆汁淤积症(1CP)患者胎盘中NPY在各临床分度中的表达及其在疾病中的临床意义。方法:将60例ICP分为轻度ICP(20例)、中度ICP(20例)、重度ICP(20例)3组,采用免疫组化方法对3组患者胎盘中NPY的表达进行定位分析,并通过计算机图象系统分析NPY表达的程度及强度,同时进行半定量分析。结果:①NPY表达于胞浆或胞膜上;②重度ICP组NPY胎盘中的表达明显高于中度ICP组与轻度ICP组,图象分析NPY在轻度ICP组、中度ICP组、重度ICP组的平均光密度值(OD值)分别为(0.9230±0.1524)、(1.0738±0.1493)、(1.6003±0.1182),统计学分析:轻度、中度组之间P>0.05,重度、轻度组之间P<0.01,重度、中度组之间P<0.01。结论:NPY表达水平与ICP的临床分度、疾病严重程度、发生发展、预后有关。

Objective： To investigate the expression of NPY on the placenta of ICP in various clinic stages and their clinical significance. Methods： The placentas of 60 ICP （mild 20, moderate 20, severe 20） women were used for detecting expression and quantification of NPY by immunohistochemistry staining and image analysis. The expression levels of NPY among groups were compared. Results： （ 1 ） NPY protein was expressed in cell plasma. （2） The expression level of NPY was significantly higher in severe ICP group than that in the other two groups, its expression was significantly different between the severe and mild ICP （P 〈 0. 01 ） . Furthermore, a significant difference between the moderate and severe ICP was found （ P 〈 0. 01 ） . Average value of optical density （OD） of NPY by image analysis in mild, moderate, and severe ICP was （0. 923 0 ± 0. 154 2）, （ 1. 073 8 ± 0. 149 3）, （ 1. 600 3 ±0. 118 2）, respectively. Conclusion： The expression level of NPY are correlated to the clinic stage and severity of the disease process.
