摘要
目的:建立基于胶体金免疫层析法(immuno-chromatographic assay,ICA)的敏感、特异且使用方便的嗜肺军团菌抗原检测技术。方法:采用柠檬酸盐还原法制备胶体金颗粒、标记抗嗜肺军团菌血清1型抗原的多克隆抗体,制成免疫层析检测试剂。结果:应用嗜肺军团菌ICA检测试剂可特异性地检测出嗜肺军团菌血清1型抗原,最低检出量为10 ng/ml,与其他血清型抗原不发生交叉反应,与美国Binax NOW ICT试剂盒的检测结果一致。结论:嗜肺军团菌ICA检测试剂具有特异性强、敏感性高、简便快速的特点,适合基层单位和现场使用。
Objective: To develop a method of gold-immunochromatography assay (ICA) with sensitive, specific, rapid and simple characteristics for the detection of Legionella pneumophila antigen. Methods: The colloidal gold labeled with the antibody of L. pneumophila serogroup 1 was prepared, and the dipstick of ICA was develvped. Results: The testing results showed that only L. pneumophila serogroup 1 antigen was detected, its limit of detection being 10 ng/ml and conforming to the Binax NOW Legionella Urinary Antigen Test. Conclusion: ICA test is a specific ,sensitive,simple and rapid assay for detecting L. pneumophila antigen.
