摘要
目的观察银杏叶提取物(舒血宁注射液)对高脂饮食诱导的胰岛素抵抗(IR)大鼠血清瘦素、脂联素及肿瘤坏死因子-α(TNF-α)、游离脂肪酸(FFA)的影响。方法采用高脂喂养的方法建立IR大鼠动物模型,舒血宁腹腔注射进行干预,罗格列酮作为阳性对照药物。干预前后分别行高胰岛素-正葡萄糖钳夹实验测定葡萄糖输注率(GIR),并检测空腹血糖(FBG)、空腹胰岛素(FINS)、血清FFA、TNF-α、瘦素、脂联素等指标。结果高脂喂养成功复制了IR大鼠模型,第4周即出现GIR下降,随时间延长,GIR下降更加明显,与正常对照组相比,差异有统计学意义(P<0.01);在胰岛素敏感性下降同时,高脂对照组大鼠还表现为内脏脂肪相对含量及血清TNF-α、FFA升高,脂联素降低;银杏叶提取物干预不仅使葡萄糖输注率明显改善(P<0.01),而且在一定程度上降低TNF-α和FFA(P<0.05),升高脂联素(P<0.01),与罗格列酮作用相近。各组大鼠血清瘦素水平无显著差异。GIR与FFA呈显著负相关,与脂联素呈显著正相关。结论银杏叶提取物可以调节TNF-α、FFA、脂联素等脂肪细胞因子的分泌,这种调节与胰岛素敏感性的改善作用有着一定的联系。
Objective To observe the effect of ginkgo biloba extract (EGb) on the serum leptin, adiponectin, TNF-α and FFA concentration in insulin resistant rats induced by high fat diet. Methods Insulin resistant rats were induced by high fat diets firstly, and from the 4th week, the rats were intervened by EGb or rosiglitazone respectively. The intervention lasted for four weeks. Before and after the intervention, the hyperinsulinemic-euglycemic clamping was performed to determine the glucose infusion rate (GIR) and blood samples were taken to be detected fasting blood glucose, fasting insulin, free fatty acid, tumor necrosis factor-α, leptin and adiponectin. Results The insulin resistant model was successfully duplicated by high fat feeding. At the 4th week, the GIR decreased;and at the 8th week, the GIR decreased more significantly. There was statistical difference between the high fat feeding group and the normal control group (P 〈 0.01 ). The high fat feeding group also showed high serum TNF-α and FFA, hypoadiponectinimia, the content of visceral fat tissue increased significantly. EGb not only increased GIR, but also decreased the level of serum TNF-α and FFA, increased the level of adiponectin, and the effect was similar with rosiglitazone. There was no difference in leptin level among these groups. The correlation analyze showed FFA was negatively correlated with GIR and the adiponectin was positively correlated with GIR. Conclusion EGb can modulate the secretion of adipocytokine, such as TNF-α, FFA and adiponectin. The modulation has correlation with the improved insulin sensitivity.
