银莱汤对肺胃积热证小鼠血清TNF-α和IL-2的影响 被引量:12

Influence of Yinlai Decoction on serum IL-2 and TNF-α levels in mice with lung-stomach heat retention syndrome

摘要 目的制作阳明积热证、肺胃积热证中医证候模型;探讨银莱汤对肺胃积热证防治作用的机理;证明肺胃同治理论的有效性,提倡治疗肺热证重视阳明的思路。方法先予小鼠高热量高蛋白饲料以制作阳明积热证模型,在此基础上,经鼻感染FM1甲型流感病毒,制作肺胃积热证模型,分别予小儿消积止咳口服液、利巴韦林及高、低剂量银莱汤灌胃,比较各组血清白细胞介素(IL-2)和肿瘤坏死因子(TNF-α)水平。结果阳明积热证小鼠胃电图与正常组比较呈高频高幅(P<0.05);利巴韦林组、银莱汤低剂量组血清TNF-α水平低于肺胃积热证组(P<0.01),银莱汤高、低剂量组血清TNF-α水平与小儿消积止咳口服液组和利巴韦林组无显著性差异(P>0.05);小儿消积止咳口服液组、利巴韦林组、银莱汤高剂量组血清IL-2水平均高于正常组、肺胃积热证组(P<0.01)。结论银莱汤能够明显改善肺胃积热证小鼠的症状体征,并可抑制肺胃积热证小鼠血清TNF-α释放,促进血清IL-2释放,说明银莱汤可以调节机体免疫功能、对抗流感病毒导致的炎症损伤,论证了治疗肺热证重视阳明的有效性。 Objective To establish the mouse model with yangming heat retention syndrome and lung- stomach heat retention syndrome and to study the therapeutic mechanism of Yinlai Decoction treating mice with lung-stomach heat retention syndrome, then to demonstrate the effectivity of treating lung and stomach simultaneously. Methods Mouse model of lung-stomach heat retention syndrome was established by feeding high calorie and protein diet, and by infecting FM1 influenza virus via nose. Five groups, with 10 mice in each group,were formed and two dosages of Yinlai Decoction, saline, Xiaoerxiaojizhike Oral Liquid, and ribavirin were administrated for 7 days. The lung pathological damage, serum IL-2 and TNF- α levels were measured. Results EGG (electro gastrography) frequency and swing were higher in the mouse model of lung-stomach heat retention syndrome than those in the normal mice ( P 〈 0.05 ). Levels of serum TNF-α were significant lower in ribavirin and low dosage of Yinlai Decoction group than those in the mouse model group (P 〈 0.01 ). There were no significant differences in levels of serum TNF-α between Yinlai Decoction group and Xiaoerxiaojizhike Oral Liquid or ribavirin group ( P 〉 0.05 ). Levels of serum IL-2 were higher in high dosage of Yinlai Decoction, ribavirin and Xiaoerxiaojizhike Oral Liquid group than those in the saline and model group(P 〈 0.01 ). Conclusion Yinlai Decoction may improve the symptoms and objective signs of mice with lung-stomach heat retention syndrome, and decrease the levels of serum TNF-α and increase the levels of serum IL-2. Therefore, Yinlai Decoction may regulate the immune function to relieve the injury of lungs caused by influenza virus infection. It is demonstrated that attaching importance to yangming is effective when treating lung heat syndrome.
