摘要
目的:建立等度高效液相色谱法(HPLC)分离测定保健食品中大豆异黄酮三种苷及其苷元的方法。方法:采用Inert-silODS-3(4.6mm×150mm,5μm)色谱柱,以1.5%乙酸-甲醇-乙腈(70∶22.5∶7.5)为流动相等度洗脱,检测波长260nm,柱温55℃,流速1.2mL·min-1。样品用70%的乙醇超声提取,经0.45μm滤膜过滤后进样。结果:HPLC过程于30min左右完成,大豆苷和黄豆苷的分离度为0.9以上,余者分离度均大于1.5。对4种市售保健食品作了分析。结论:方法简便、快速、准确,为含大豆异黄酮的保健食品含量测定及质量控制提供了较为理想的方法。
Objective:To establish a method for the determination of soybean isoflavones including throe glycosides and their aglycones in health-care foodstuffs by high performance liquid chromatography (HPLC) with isocratic elution. Methods:The HPLC system consisted of Inertsil ODS-3 column (4. 6 mm×150 mm, 5μm) , column temperature at 55 ℃, with methanol-acetonitrile -1.5% acetic acid (22. 5:7.5:70) as mobile phase. Detection wavelength was at 260 nm. Samples were extracted by 70% ethanol with ultrasonic wave, and then filtrated with 0. 45 μm filtration membrane. Results: The separate rate of daidzin and glycitin was 1.0, and the others were all higher than 1.5. Each determination was finished within 30 minutes. Four products from the market were analyzed by this method. Conclusion: The method is simple, rapid and applicable for the determination of soybean isoflavones, and it provides a pref- erable assay method for the quality control of these health-care foodstuffs containing soybean isoflavones.
