摘要
目的观察联合应用GM1和NGF时大鼠面神经核内神经元p75NGFR的表达情况,探讨GM1提高NGF在促进运动神经元再生方面的作用机制。方法健康成年SD大鼠24只,随机分为2组,每组12只:四周组和八周组。利用大鼠面神经再生室模型和免疫组化方法,结合细胞计数和灰度分析的手段,对大鼠面神经核内p75NGFR免疫阳性神经元进行观察。结果四周组和八周组的实验侧的p75NGFR表达阳性细胞数明显高于对照侧(P<0.01);四周组的p75NGFR阳性细胞数明显高于八周组(P<0.01)。面神经核p75NGFR阳性细胞灰度值无论四周组和八周组之间,还是实验侧与对照侧之间都没有显著性差异(P>0.05)。结论GM1可以通过抑制面神经核内p75NGFR阳性神经元的凋亡的机制强化NGF促进神经再生和成熟的作用。
Objective To explore the mechanism underlying the regeneration of motor nerves by NGF and GM1. Methods Twenty four adult SD rats were divided randomly and equally into 4 - week and 8 - week group. Immunohistochcmistry for p75NGFR in nucleus of facial nerves was performed with cells counting and gray level analysis. Results The neurons expressed p75NGFR of the experimental sides were much more than that of the control sides in both 4 -week and 8 -week group ( P 〈0.01). The neurons of both sides in4 -week group were also much more than that in 8 - week group ( P 〈 0.01 ). However, the gray level of the neurons expressed p75NGFR were not different among the groups and the sides ( P 〉 0. 05 ). Conclusion GM1 can enhance the promotion of NGF on the regeneration of motor nerves through the mechanism that GM1 prevents neurons in nucleus of facial nerves which express p75NGFR from languishment.
