摘要
目的:研究系膜增生性肾炎(MsPGN)肾小球系膜细胞(GMC)凋亡和尿表皮生长因子(EGF)的变化特征,探讨慢肾蛋白停对其的影响及作用机制。方法:采用手术摘除一侧肾脏加腹腔多次注射BSA与LPS及皮下多次注射完全弗氏佐剂,复制MsPGN动物模型。慢肾蛋白停予以防治性治疗,放免法测定尿EGF含量,TUNEL法检测GMC凋亡现象。结果:模型组GMC凋亡率降低,尿EGF含量增高;治疗组GMC凋亡率增高,尿EGF水平下降。结论:慢肾蛋白停可以调节肾内EGF的异常分泌状态,促进GMC凋亡、抑制GMC增生和基质的增多,减轻肾脏组织的病理损害。
Objective: To observe the changes of mesangial cells apoptosis and urinary epidermal growth factor in mesangial proliferative glomerulonephritis(MsPGN) rats , the effects of Chinese medicine manshen danbai ting and discuss its mechanism. Methods: The models with MsPGN were made by extracting one kidney and intraperitoneal injecting BSA,LPS and subcutaneous injecting completed Freund's adjuvant. Rats were divided into four groups: normal, model and two medicine groups, meanwhile corresponding medicines were given. EGF content was tested by radioimmunoassay, the apoptosis of GMC was determined by Tunel. Results: The EGF level in model group was higher than that in the normal, the apoptosis rate of mesangial cells was significantly lower. Manshen danbai ting could lower the EGF level in rats with MsPGN and elevate the apoptosis rate of mesangial cells, there were obviuos differences between the two groups.(P<0.05). Conclusion: Manshen danbai ting can inhibit the proliferation of mesangial cells and matrixes, relieve the kidney injury by regulating the EGF abnormal secretion and promoting the apoptosis of mesangial cells.
