摘要
目的探讨胃癌组织中HIF-1α、Glut1及增殖细胞核抗原(PCNA)蛋白的表达及其临床意义。方法采用免疫组织化学染色法检测HIF-1α、Glut1及PCNA在52例胃癌组织及20例正常胃黏膜的表达水平,并分析其与胃癌生物学行为的关系。结果HIF-1α、Glut1在正常胃黏膜均无表达,胃癌组织中的阳性率分别为69.2℅和75℅,与正常组织比较差异有统计学意义(P<0.01)。HIF-1α蛋白阳性表达率与TNM分期和淋巴结转移有关,差异有统计学意义(P<0.01);Glut1蛋白阳性表达率与分化程度、TNM分期和淋巴结转移有关,差异有统计学意义(P<0.01);HIF-1α与Glut1表达呈正相关(r1=0.580,P<0.01);PCNA在胃癌中的表达(65.19%±20.82%)显著高于正常组织中的表达(15.50%±9.72%)(P<0.01)。HIF-1α、Glut1蛋白的异常表达与PCNA呈正相关(r2=0.723,P<0.01;r3=0.527,P<0.01)。结论胃癌组织HIF-1α、Glut1、PCNA蛋白过度表达在胃癌的发生、发展过程中可能具有协同作用,对于胃癌的诊断及预后判断有一定的指导意义。
Objective To explore expressions of HIF-1α,Glutl and PCNA in gastric cancer, and to analyze the clinical significance. Methods Immunohistochemical staining method was used to detect expressions of HIF-1α, Glutl and PCNA in 52 specimens of gastric cancer and 20 specimens of normal gastric tissues. Furthermore,their interrelations with biological behaviour of gastric cancer were analyzed. Results None of the normal gastric tissues expressed HIF-1α and Glut1. Whereas positive expression rates of HIF-1 et and Glutl in gastric cancer were 69.2 % and 75% ,respectively. HIF-1α expression correlated with TNM stage(P 〈0.01 ),lymph node metastasis(P 〈0.01 ) and Glutl expression ( r1 = 0. 580 ,P 〈 0.01 ). Glut1 expression correlated with cell differentiation ( P 〈 0.01 ) , TNM stage ( P 〈 0.01 ) and lymph node metastasis(P 〈0.01 ). PCNA level was higher in gastric cancer(65.19% ± 20.82% ) than that in normal gastric tissues( 15.50% ± 9.72% ) ( P 〈 0.01 ). The positive expressions of HIF-1 et and Glutl were positively correlated with PCNA level( r2 = 0. 723, P 〈 0.01 ; r3 = 0. 527, P 〈 0.01 ). Conclusion The overexpressions of HIF-1 et, Glutl and PCNA protein are closely associated with carcinogenesis and progression of gastric cancer. And it is of clinical signifi- cance to guide the diagnosis and prognosis of gastric cancer.
