肝癌组织FHIT和Survivin表达及其相关性的探讨 被引量:3

摘要 目的:分析脆性组氨酸三联体(fragilehistidine triade,FHIT)基因和生存素(Survivin)在原发性肝细胞性肝癌(pri mary human hepato-cellular carcinoma,HCC)中的表达及探讨两者之间的关系。方法:分别应用免疫组化SP法和原位杂交的方法检测HCC50例、肝硬化30例、正常肝组织10例中FHIT蛋白、Survivin蛋白和FHIT mRNA、Survivin mRNA的表达情况。结果:HCC、肝硬化和正常肝组织的FHITmRNA、FHIT蛋白阳性表达率差异有统计学意义(χ2=15.74,χ2=15.74,P均<0.05),FHITmRNA在HCC组织中的表达明显低于在肝硬化和正常肝组织中的表达(χ2=13.45,χ2=7.55,P均<0.05),FHIT蛋白在HCC组织中的表达明显低于在肝硬化和正常肝组织中的表达(χ2=12.52,χ2=5.42,P均<0.05)。HCC、肝硬化和正常肝组织的Survivin mRNA、Sur-vivin蛋白阳性表达率不同,三者之间的差异有统计学意义(χ2=19.59,χ2=28.44,P均<0.05),Survivin mRNA在HCC组织中的表达明显高于在肝硬化和正常肝组织中的表达(χ2=10.25,χ2=12.50,P均<0.05),Survivin蛋白在HCC组织中的表达明显高于在肝硬化和正常肝组织中的表达(χ2=21.19,χ2=19.47,P<0.05)。结论:FHIT、Survivin在HCC组织中的表达关系密切,呈负相关;FHIT、Sur-vivin在HCC的发生、发展过程中起着重要的作用,联合检测可为HCC的早期诊断、进一步治疗及预后判断提供必要的理论依据。 OBJECTIVE: To explore the expressions of FHIT and Survivin in primary human hepatocellular carcinoma and study the relationship between them. METHODS: The expression of FHIT protein and Survivin protein in 50 HCC, 30 cirrhosis and 10 normal tissues were assessed by the immunohistochemical method. The expressions of FHIT mRNA and Survivin mRNA in 50 HCC, 30 cirrhosis and 10 normal tissues were assessed by in situ hybridization. RESULTS: The rates of expressions of FHIT mRNA and FHIT protein in cancer tissues, cirrhosis tissues and normal tissues were significantly different (X^2 = 15.74, X^2 = 15.74,P〈0.05). The rate of expression of FHIT mRNA in cancer tissues was lower than those in cirrhosis tissues and normal tissues (X^2 = 13.45, X^2 =7.55, P〈0.05). The rate of expression of FHIT protein in cancer tissues was lower than those in cirrhosis tissues and normal tissues (X^2 = 12.52 ,X^2 = 5.42, P〈0.05). The rate of expressions of Survivin mRNA and Survivin protein in cancer tissues, cirrhosis tissues and normal tissues were significantly different (X^2 = 19.59, X^2 = 28.44,P〈0.05). The rate of expression of Survivin mRNA in cancer tissues was higher than those in cirrhosis tissues and normal tissues (X^2=10.25,X^2=12.50, P〈0.05). The rate of expression of Survivin protein in cancer tissues was higher than those in cirrhosis tissues and normal tissues (X^2=21.19,X^2= 19. 47,P〈0.05). CONCLUSIONS: The expression of FHIT is closely associated with the expression of Survivin in HCC and they take negative correlation. FHIT and Survivin may play important roles in oneogenesis and progression of HCC. Detection of FHIT and Survivin may have potential values on early diagnosis, treatment and predicting the prognosis of HCC.
