摘要
目的:研究不同侵袭力癌细胞的内质网膜系统(endoplasmic reticulum,ER)结构及分布特点。方法:利用常规电镜固定方法及高锰酸钾技术,在透射电镜(TEM)和扫描电镜(SEM)下对侵袭力有明显差异的两种人大肠癌细胞ER的超微结构进行了观察和研究。结果:高侵袭力的CCL229细胞异型明显,细胞伸出的大量形态各异的伪足中含有丰富的小泡状和扁平囊样rER。细胞质中ER含量亦比较丰富,核周ER致密并在观察中首次发现由众多小泡组成的链珠状"膜流"结构从核周一直深入到伪足中。
Methods: Comparative studies of the endoplasmic reticulum (ER) in two colorectal carcinoma cell lines were performed in the routine TEM and SEM with potassium permanganate fixation. Results: The more invasive CCL229 cells showed obvious changes with rich ER, throughout cytosol and in many pseudopodia or cell processes, but had less Golgi complex. Membranous flow-like structures from perinuclear region to pseudopodia were observed also in CCL229 cells.
