脑缺血大鼠脑内Ⅱ型磷脂酶A_2 mRNA表达

摘要 采用改良Zieve法及RNA斑点杂交技术,分别检测了大脑中动脉阻塞后大鼠脑内磷脂酶A2(PLA2)活力及Ⅱ型PLA2mRNA表达的变化,旨在了解脑缺血后脑内PLA2活力的变化及参与脑缺血病理过程的PLA2类型。研究发现:与假手术组相比,脑缺血组大鼠脑内PLA2活力显著增高(P<0.05);假手术对照组大鼠脑内Ⅱ型PLA2mRNA表达水平很低,脑缺血组大鼠脑内Ⅱ型PLA2mRNA表达明显增强。结果提示:Ⅱ型PLA2参与了脑缺血病理过程,有必要进一步研究Ⅱ型PLA2mRNA表达抑制剂对脑缺血的保护作用。 In order to study if,and which group of phospholipase A 2(PLA 2) is involved in brain ischemia,the improved Zieve method and RNA dot blotting technology were adopted to examine changes of PLA 2 activity and expression of group ⅡPLA 2 mRNA in ischemic brain in rat.The brain ischemia model was produced by occluding the left middle cerebral artery of rat for 3 hours with a varnish cylinder attached to a nylon thread inserted through the internal carotid artery.It was found that the PLA 2 activity in left forebrain in rat in brain ischemia group was increased markedly compared with that in sham control group (P<0.05).The expression of group Ⅱ PLA 2 mRNA was poorly detected in left forebrain in rat in sham control group,whereas group Ⅱ PLA 2 mRNA was abundantly expressed in brain ischemia group.These results guggest that group Ⅱ PLA 2 is involved in brain ischemia and it ,s necessary to study whether group Ⅱ PLA 2 mRNA expression inhibitor is useful in preventing ischemic brain damage.
