摘要
目的:探讨基质金属蛋白酶(MMP-2、MMP-9)在扁平苔藓皮损及外周血中的表达及意义。方法:应用免疫组化SP法对33例扁平苔藓及20例正常皮肤组织MMP-2、MMP-9的表达进行观察,进一步采用双抗体夹心酶联免疫吸附法检测22例扁平苔藓和16例正常人血清中MMP-9水平。结果:MMP-2在扁平苔藓及正常皮肤组织几乎均无免疫组化着色。MMP-9在扁平苔藓强烈表达于表皮角质形成细胞及真皮淋巴细胞胞浆,其表达显著高于正常皮肤(P<0.01)。扁平苔藓患者血清MMP-9水平明显高于对照组(P<0.01)。结论:MMP-9在扁平苔藓基底膜带降解中发挥了作用。
Objectives: To determine the expression of matrix metalloproteinases (MMP - 2 and MMP - 9 ) and to explore its significance in lichen planus (LP). Methods: The expressions of MMP- 2 and MMP-9 in 33 LP lesions and 20 normal skin specimens (controls) were measured by immunohistochemistry. The level of MMP - 9 in sere was measured by double antibody sandwich enzyme - linked immunosoibent assay (ELISA) in 22 LP patients and 16 normal controls. Results: The expression of MMP- 2 was negative in near all LP lesions and normal skin tissues. The expression of MMP- 9 was markedly increased in cytoplasm of keratinocytes and dermal inflammatory infiltrates in LP, and significantly stronger than the expression in normal controls ( P 〈 0.01 ). Serum level of MMP- 9 was significantly higher in LP patients than in normal controls ( P 〈0.01). Conclusions: MMP- 9 may play a role in degrading the basement membrane zone of LP.
