摘要 目的评价前房插管法测量小鼠眼压的准确性并测量5种不同品系小鼠的眼压。方法选择3个月龄清洁级C3H/HeJ、C57BL/6J、BALB/CJ和ICR小鼠各20只,3、6、9、11个月龄DBA/2J小鼠各7只,用调整与小鼠玻璃体腔相连的水柱高度来检测前房插管测量系统眼压测量的准确性,将连接于前房插管测量系统的玻璃微管插入小鼠前房,测量小鼠的眼压。结果前房插管系统测得的眼压值和实际眼压值之间有很好的相关性(r=0·99,P=0·000);使用该技术测得C3H/HeJ、C57BL6J、BALB/CJ和ICR小鼠的眼压分别为(10·64±1·92)、(10·81±1·84)、(10·98±1·3)和(11·60±1·29)mmHg;3、6、9和11个月龄DBA/2J小鼠的眼压分别为(11·43±1·84)、(10·54±2·49)、(19·35±7·49)和(26·76±4·4)mmHg。结论前房插管技术可以准确地测量小鼠的眼压,3个月龄5种小鼠眼压相似,DBA/2J小鼠眼压在9个月龄和11个月龄时增高。 Objective Mouse is a particularly attractive model for glaucoma research at present. Technical difficulty associated with the measurement of intraoeular pressure (IOP) in mouse eyes has, however,limited the use of these animals in certain experimental paradigms. This study was taken to validate the accuracy of mieroneedle method and measurement of the IOP in 5 kinds of inbred mice. Methods Five different kinds of inbred mice including 20 mice of C3H/HeJ,C57BL6J,BALB/CJ and ICR and 7 DBA/2J mice were selected for the study. The age of the mice was 3,6,9 and 11 months. The animals were raised at the cycle of 14 hours of light environment and 10 hours of night environment for two weeks. The IOP of the mice was measured with a very fine fluid-filled glass mieroneedle device connecting ocular anterior chamber of mouse. The accuracy of IOP measured with the mieroneedle system was validated by adjusting the height of a water column connected to the mouse vitreous body. Results The IOP readings generated by mieroneedle device were correlated with the actual pressure in the mouse eye (r = 0.99, P =0. 000). The IOP value obtained by mieroneedle device was 10.64 ± 1.92, 10. 81 ± 1.84,10. 98 ± 1.3 and 11.60 ± 1.29 mmHg respectively in C3H/HeJ, C57BL6J, BALB/CJ and ICR mouse. At 3-,6-,9- and 11-month-old DBA/2J mouse, the IOP was 11.43 ± 1.84,10.54 ± 2.49,19.35 ±7.49 and 26. 76 ± 4. 4 mmHg, respectively. The fraction of eyes with IOP 〈 15 mmHg was 100% ,93% ,38% and 0 respectively and that with IOP 〉 20 mmHg was 0,0,38% and 91%. Conclusion Mieroneedle measurement system is an accurate method for measurement of mouse IOP. There is no significant difference of IOP in five different kinds of 3-month-old inbred mice. The IOP presents an increase value in 9-and 11-month-old DBA/2J mice.
