摘要
目的探讨地塞米松(DEX)对大鼠房水中转化生长因子β(TGF-β)及晶状体上皮细胞(LECs)中TGF-β受体表达的影响及其在抑制后发性白内障(PCO)中的作用。方法1个月龄SD大鼠眼局部应用DEX半年,用药后每月取房水,ELISA法检测TGF-β,用药1、3、6个月后,免疫组织化学法检测LECs中转化生长因子β受体(TGF-βR)的表达,RT-PCR检测LECs中TGF-βR mRNA的表达。培养大鼠LECs,不同浓度DEX干预24h,RT-PCR和Western blot法检测TGF-βR的表达。结果各时段房水中TGF-β质量浓度与正常对照组相似(P>0·05);TGF-βR于用药1、3、6个月后的表达与对照组比较均降低(P<0·05),TGF-βR mRNA在上述时段分别为1·07±0·19、0·85±0·10、0·65±0·09,对照组分别为1·53±0·15、1·08±0·13、0·87±0·15,各时段组间差异有统计学意义(P<0·05)。DEX干预培养的LECs后,TGF-βR分别为0mol/L组1·11±0·13,10-8mol/L组0·98±0·12,10-7mol/L组0·49±0·07,10-6mol/L组0·27±0·07,组内差异有统计学意义(P<0·05)。结论DEX降低LECs中TGF-βR的表达,可能是抑制PCO形成的一个重要途径。
Objective To assess the effect of dexamethasone (DEX) on transforming growth factor-β (TGF-β) in rat aqueous humor and expression of TGF-β receptor in rat lens epithelial cells (LECs) and investigate the mechanism of suppression of DEX on formation of after cataract. Methods DEX was applied topically to eyes of 1-month-old Sprague-Dawley rats for six months. Aqueous humor was collected to detect TGF-β level by ELISA at the end of 1,2,3,4,5 and 6 months after DEX therapy, respectively. The anterior capsules of lens from all rats were prepared to detect TGF-β receptor by immunofluorescence and the capsule tissues were applied to assay TGF-β receptor mRNA after 1,3 and 6 months following DEX treatment. Primary rat LECs were cultured for in vitro study. The cultured cells were exposed to different concentrations of DEX for 24 hours. RT-PCR and Western blot were applied to identity the expression of TGF-β receptor at protein and transcription levels. Results TGF-β levels in rat aqueous humor revealed no significant differences between control and DEX-treated group at various time (P 〉 0. 05). Outcome from immunofluorescence showed that expression of TGF-β receptor (IOD) down-regulated after DEX application at 1,3 and 6 months compared to control groups (P 〈 0.05 ). RT-PCR exhibited that expression of TGF-β receptor mRNA was 1.07 ± 0.19,0. 85 ± 0. 10,0.65 ± 0. 09 respectively in DEX treating for 1,3,6 months, and that in the control group was 1.53 ± 0. 15,1.08 ± 0. 13,0. 87 ± 0. 15, respectively, showing significant differences between DEX group and control group at various time (P 〈 0. 05 ). Expression of TGF-β receptor in cultured rat LECs at protein and transcription level was evidently declined in DEX group (P 〈 0. 05 ). The expression of TGF-β receptor mRNA was 1.52 ± 0.16,1.15 ± 0. 08,0.96 ± 0. 06 and 0.84 ± 0. 05 in 0,10^ -8 , 10 ^-7 and 10^ -6 mol/L concentrations of DEX groups, respectively. The expression of TGF-β receptor was 1.11 ±0.13,0.98 ±0.12,0.49 ±0.07 and 0.27 ±0.07 in 0,10^-8,10^-7 and 10^-6mol/L concentrations of DEX groups, respectively. Conclusion Inhibition of DEX on TGF-β receptor expression in LECs may play an important role in suppressing the formation of after cataract.
