摘要 目的:观察成人不同部位角质形成细胞的生物学差异,探讨用来构建自体组织工程皮肤的最佳表皮细胞来源.方法:体外分离培养成年男性头皮、躯干、下肢及包皮来源的角质形成细胞,分别利用绘制细胞生长曲线、3H-胸腺嘧啶(3H-TdR)掺入、流式细胞仪检测等方法比较细胞在生长增殖、细胞周期及代谢等方面的生物学差异.结果:在细胞的生长增殖及DNA合成代谢方面,头皮与包皮来源的角质形成细胞之间无统计学差异(P>0.05),躯干与肢体来源的细胞之间亦无统计学差异(P>0.05),但前两者均高于后两者(P<0.05);细胞周期分析表明,头皮及包皮来源的角质形成细胞S期比例高于躯干及肢体组.结论:头皮及包皮组织来源的角质形成细胞可作为构建自体组织工程皮肤的表皮细胞来源. AIM: To investigate the biological characteristics of keratinocytes derived from different parts of the human skin and discuss an optimal seed for the construction of tissue engineered skin. METHODS: The human kerafinocytes were harvested from the different parts of human male skin such as foreskin, scalp, trunk and limbs ( aged 25 - 35 years ). The keratinocytes were cultured in K-SFM serum free culture medium in vitro. The growth kinetic characteristics were studied by cell growth curve and the 3 H-TdR incorporation was employed to measure the DNA metabolism. The cell cycle was determined with flow cytometry. RESULTS: For the DNA metabolism and cell proliferation, there was no clear difference between scalp and foreskin groups (P 〉 0.05 ). The same results were got from trunk and limbs groups ( P 〉 0.05 ). The difference was significant between the groups of scalp and foreskin and the groups of trunk and limbs ( P 〈 0.05 ). The percentage of cells in S phase of scalp and foreskin groups was much higher than that of trunk and limbs groups. CONCLUSION: It might be better to take the keratinocytes from scalp or foreskin as the seeding cells to construct the tissue engineered skin.
