抑制NF-κB对PDGF-A、PDGF-B在糖尿病大鼠肾组织表达的影响 被引量:1

摘要 目的:研究糖尿病大鼠肾组织血小板源性生长因子-A(PDGF-A)、血小板源性生长因子-B(PDGF-B)、核因子-κB(NF-κB)的表达,以及NF-κB抑制剂PDTC对其表达的影响。方法:将30只健康Wistar雄性大鼠随机分成3组:正常对照组(NC组)、糖尿病组(DM组)和糖尿病PDTC干预组(DP组),每组各10只。8周末收集大鼠24h尿测定微量白蛋白,处死大鼠取出肾脏称重,计算尿微量白蛋白排泄率(UAER)、肾重指数(KWI)并用免疫组化方法测定PDGF-A、PDGF-B、NF-κB的含量。结果:8周末,DM组和DP组的UAER、KWI较NC组明显升高;DP组较DM组明显下降。DM组肾组织NF-κB和PDGF-B的表达较NC组显著增高,DP组较DM组明显下降,但显著高于NC组;三组大鼠肾组织PDGF-A的表达均无显著性差异。结论:糖尿病大鼠肾组织局部表达的PDGF-B、NF-κB明显增加,抑制NF-κB能明显降低PDGF-B表达。提示NF-κB和PDGF-B可能在糖尿病肾病发生发展中起重要作用。 Objective: To investigate the expressions of PDGF-A, PDGF-B and NF-κB in the renal of diabetic rats, and their changes when the expression of nuclear factor-kappa B (NF-κB) was inhibited by PDTC. Methods: Thirty healthy male Wistar rats were divided randomly into three groups as normal control group (NC group), diabetic model group (DM group) and diabetic rats with treatment of PDTC (DP group) with 10 rats in each group. Eight weeks later, UAER was determined, the rat's kidney was weighed and kidney weight index (KWI) was calculated. The expressions of PDGF-A, PDGF-B and NF-κB were observed by immunhistochemistry staining. Results: At the end of the 8th week, UAER and KWI in the group DM and DP were significantly higher than those in the NC group, and both decreased significantly in the DP group. The expression of PDGF-B and NF-κB in the DM group were significantly higher than those in the NC group, the expression of those in the DP group decreased significantly than those in the DM group, but significantly higher than those in the NC group. The expression of PDGF-A was not significantly different among the three tissue may be involved in t the NF-κB expression can groups. Conclusion:NF-κB and PDGF-B expressed locally in the renal he beginning and progressing of diabetic nephropathy, and inhibition of decrease the expression of PDGF-B.
