色素上皮衍生因子在糖尿病大鼠肾脏中表达的改变

摘要 目的:探讨糖尿病大鼠肾脏色素上皮衍生因子(PEDF)的表达。方法:将40只雄性Wistar大鼠随机分为:正常对照组(NC)、糖尿病模型组(DM)。分别于4周、8周末,检测血糖、糖化血红蛋白、血脂、肾重指数、尿白蛋白排泄率,应用免疫组织化学染色法检测肾脏PEDF的表达。结果:4周、8周末DM组肾重指数、尿白蛋白排泄率均明显高于NC组(P<0.01);两组血脂无明显差别(P>0.05);DM组大鼠肾脏PEDF的表达较NC组明显减少(P<0.01);PEDF的表达与肾重指数、尿白蛋白排泄率均呈明显负相关(r=-0.896,P<0.01;r=-0.884,P<0.01)。结论:肾脏PEDF表达不足可能是糖尿病肾病发生发展的主要原因之一。 Objective. To investigate the expression of pigment epithelium-derived factor (PEDF) in kidneys of diabetic rats. Methods: Forty male Wistar rats were randomly divided into two groups as normal control(NC) group and diabetic model(DM) group. At the end of the 4th week and 8th week, the blood glucose, glycosylated hemoglobin, plasma lipid, kidney weight index and urinary albumin excretion rate were measured. At the same time, the expression of PEDF in renal was detected by immunohistochemistry staining. Results: At the end of 4th week and 8th week, compared with those in NC group, kidney weight index and urinary albumin excretion were signifi- cantly higher in DM group(P〈0.01). However, the level of plasma lipid in DM group and NC group had no significant difference(P〉0.05). Compared with that in NC group, the expression of PEDF was significantly lower in the kidney of DM group(P〈0.01). The expression of PEDF showed a strong negative correlation with kidney weight index and urinary albumin excretion rate (r=-0. 896, P〈0.01; r= -0. 884, P〈0.01). Conclusion: The down-regulation of PEDF may be one of the main reasons for the onset and development of diabetic nephropathy.
