摘要
以麝香百合幼嫩花部组织为外植体进行离体培养,研究了不同部位外植体和植物生长调节物质对其愈伤组织的诱导及植株再生的影响。结果表明,不同外植体诱导愈伤组织的能力不同,由强到弱依次为花丝>花托>花梗>花柱>花瓣。在愈伤组织诱导过程中,花丝、花托及花梗在培养基MS+NAA 1.0 mg.L-1(单位下同)+BA 0.5上的诱导率都高达80%以上。交替在MS+NAA 1.0+BA0.5和MS+NAA 0.5+BA 0.25两种培养上培养最有利于愈伤组织的增殖。不定芽的分化以MS+KT 1.0+NAA 0.5最佳。生根培养基以MS+NAA 0.2效果为好。
Using the filaments, receptacles, pedicels, ovaries and petals of Lilium longiflorum Thunb.as the explants to in vitro culture, the effect of inducing calli and plant regeneration of different explants and different plant regulators was studied in this paper. The result indicared that the ability Of inducing calli of different explants was different, filaments〉receptacles〉 pedicels〉 styles〉 petals. The inducing rates of filaments, receptacles and pedicels were over 80% while culturing on MS mediums supplemented with BA 1.0 mg· L^-1 (the same unit as below) and NAA 0.5. The calli being cultured on MS plus BA 0.5 and NAA 1.0 and MS plus BA 0.25 and NAA 0.5 alternatively was helpful for proliferation of calli. Roots could formed on MS supplemented with NAA 0.2.
