孟士德薰衣草愈伤组织诱导和植株再生 被引量:7

摘要 取孟士德薰衣草叶片、茎段、芽为外植体,设置6组不同激素水平进行组织培养试验,结果显示:以MS+BA 2.0 mg.L-1(单位下同)+NAA 0.1或MS+BA 2.0+IBA 0.15为培养基诱导愈伤组织效果好;茎段的愈伤诱导率高达92.5%;芽的萌动早、愈伤诱导率达80%;叶的诱导效果最差,愈伤诱导率为35%,且极易褐变死亡。愈伤组织在MS+BA 1.0+IBA 0.1培养基上继续培养,形成大量的丛生芽。单个芽在1/2MS+NAA 0.2培养基上进行生根培养,生根率达98%。试验表明芽适合作快速繁殖材料,茎段适合先形成愈伤组织再诱导植株再生。 Using the leaf, the stem and the bud of L. angustifolia Munstead as explants, we carried out the tissue culture experiments with six culture medium using different levels of hormones, h was shown that the induction results of the MS+BA 2.0 mg·L^-1(the same unit as below)+NAA 0.1 or MS+ BA 2.0 +IBA 0.15 medium groups were better than the other groups .The induction rate of callus on stem reached as high as 92.5% ; The buds bud earlier, and induction rate of callus reached 80%. However, the induction effect of leaf's callus is not good, with the ratio being 35%, and turning brown death easily. Cultivated in MS+ BA 1.0 +IBA 0.1 culture medium sequentially, Subcultures bud formed massively. The rooting rate of these single bud in 1/2MS+NAA 0.2 culture medium reached 98%. These results indicated that the L.angustifolia Munstead's bud is right material for rapid propagation, while the stem should be used for inducing the plantlet regeneration after the callus formed.
