磁小体对大鼠成纤维细胞的生长抑制研究

摘要 目的:探讨磁小体对大鼠成纤维细胞的生长抑制作用。方法:通过培养大鼠L929细胞,并在培养液中加入0.1mg/ml,0.5mg/ml,1.0mg/ml,1.3mg/ml不同浓度的磁小体继续培养,观察作用24、48、72h细胞生长情况,通过MTT检测细胞吸光度,判断活细胞的含量,从而判断磁小体对细胞的生长抑制作用。结果:由MTT法得到细胞生长抑制率,除有一组达到7.97%外,其余皆为负值。结论:实验结果表明磁小体对细胞生长没有明显的抑制作用。抑制率出现负值的原因可能与磁小体最外层脂质成分与MTT和DMSO发生反应,被DMSO溶解从而产生吸光度有关。 Objective: To discuss the inhibitory effect of the magnetosome on the growth of rat fibroblasts. Methods. The L929 cell was experimentally cultured, and 0. img/ml, 0. 5mg/ml, 1.0mg/ml and 1. 3mg/ml of magnetosomes were added in the cultures to continue to culture. The growth condition of the living cells acted on for 24,48 and 72 hours was observed. The absorptivity of the cells was tested with MTT so as to detect the amount of living ceils and deter mine the inhibitory effect of the magnetosome on the cell growth. Results:The inhibitory rates of cell growth were negative except one group(7. 97%). Conclusion, The magnetosome does not have remarkable inhibitory effect On the cell growth. The negative inhibitory rate results from that the outer lipid of magnetosome may react with MTT and DMSO and be dissolved by DMSO, thus the absorptivity develops.
