摘要
目的研究Nras基因第12位密码子突变与原发性肝癌(HCC)发生发展的关系.方法采用聚合酶链延伸反应限制性片段长度多态性分析法(PCR-RFLP)对30例HCC手术切除标本Nras第12密码子点突变进行检测.结果在30例HCC中,发现Nras第12密码子点突变5例,突变率为167%.点突变与肿瘤大小、分化程度、有无转移及HBV感染无显著相关(P<005).结论N-ras基因第12位密码子突变参与了部分HCC的发生与发展.
AIM To study the role of oncogene N ras mutation in the progression of primary hepatocellular cancer (HCC). METHODS Polymerase chain reaction and restriction fragment length polymorphims (PCR RFLP) analysis were performed in 30 surgically resected HCC specimens to determine the point mutations at codon 12 of N ras gene. RESULTS Point mutation was detected in 5 of 30 (16 7%) of HCC specimens. No significant correlation was found between the positivity of N ras mutation and the status of background lesions, tumor size, differentiation, metastasis and hepatitis B virus ( P >0 05). CONCLUSION The point mutation at codon 12 of N ras gene is involved in the development of HCC.
