广西野生银星竹鼠体内寄生马尔尼菲青霉菌微卫星多态性研究 被引量:6

摘要 目的了解广西地区野生银星竹鼠携带马尔尼菲青霉菌(PM)基因多态性。方法用微卫星标记多态性分析法对40株从广西不同地区野生银星竹鼠体内寄生马尔尼菲青霉菌进行基因多态性分析。结果40只野生银星竹鼠自然携带PM阳性率100%。微卫星标记多态性分析法以高区分度(0.992)把40株PM分为22个不同的等位基因型。微卫星引物Ⅰ,Ⅱ,Ⅲ分别检测到13,9和6个等位基因。广西银星竹鼠体内寄生马尔尼菲青霉菌基因型共分为5类;最常见的是3类,共15株(37.5%)。桂中地区以1类为主,10株中占7株(70%);桂北地区以2类为主,9株中占7株(77.8%);桂南地区以3类为主,5株中占4株(80%);而桂西地区9株全部属于3类;4类和5类仅出现在桂东地区,7株中分别为6株和1株(分别占85.7%和14.3%)。结论广西银星竹鼠自然携带之马尔尼菲青霉菌的基因多态性可能存在地域上的差异。 Objective To investigate the genetic polymorphism of Penieillium marneffei in Rhizomys pruinosus, a species of bamboo rats of Guangxi Provinee of China. Methods Forty Rhizomys pruinosus in various districts of Guangxi were trapped. The internal organs (liver, kidneys and lungs) of Rhizomys pruinosus were taken and euhured on Sabouraud dextrose agar. The isolated organisms were identified as Penieillium marneffei based on the temperature-dependent diomorphology characteristics. Polymorphie mierosatellite marker (PMM) analysis was used to study the genetic polymorphism. The relationship of the typing and the geography distribution was analyzed. Results The natural carrier rate of Penieillium marneffei in 40 Rhizomys prninosus was 100%. Polymorphie mierosatellite marker analysis detected a total of 22 different allelie types for 40 isolates of PM with a high diseriminatory power ( D = 0. 992). Mierosatellites Ⅰ , Ⅱand Ⅲ detected 13,9 and 6 alleles, respectively. Five PMM dusters were identified for 4 0 isolates of PM obtained from 4 0 Rhizomys pruinosus. The most common was cluster 3 , a total of 1 5 isolates (37.5%). The common duster in the centre,north and south of Guangxi was duster 1 (7 of 10 isolates,70% ) ,duster 2 (7 of 9 isolates,77.8% ) and duster 3 (4 of 5 isolates,80% ) respectively. However,all the 9 isolates PM from the west of Guangxi belonged to cluster 3, duster 4 (6 of 7 isolates ,85.7% ) and duster 5 (1 of 7 isolates, 14.3% ) were observed only for isolates from the east of Guangxi. Conclusion There was the possible geographical genetic polymorphism in the PM from the Guangxi Rhizomys pruinosus.
