Effect of zoledronic acid on vasculogenic mimicry in osteosarcoma cell line LM8 in vitro

摘要 目的研究唑来膦酸对骨肉瘤细胞体外血管生成拟态的影响。方法应用Ⅰ型胶原蛋白凝胶的三维培养基对骨肉瘤细胞进行培养,观察骨肉瘤细胞是否能形成血管拟态以及唑来膦酸对其的影响,并运用电镜和激光共聚焦显微镜观察细胞的超微结构的改变。结果唑来膦酸明显抑制骨肉瘤细胞形成血管生成拟态,减少骨肉瘤细胞表面的微绒毛和破坏细胞骨架F-actin。结论唑来膦酸显著改变骨肉瘤细胞的超微结构,这个可能是其抑制血管生成拟态的形成的机制之一。 Objective To investigate the effect of zoledronic acid on vasculogenic mimicry formation in osteosarcoma cell line LM8 in vitro. Methods Osteosarcoma cell line LM8 was tested for their ability to form vasculogenic mimicry and the effect of zoledronic acid on vasculogenic mimicry formation in three-dimensional culture containing type I collagen. The ultrastructure of LM8 cells was determined by electron microscope and laser scanning confocal microscope. Results Zoledronic acid significantly inhibited vasculogenic mimicry formation of LM8 cells in vitro, and decreased the surface microvillus and destroyed cytoskeletal protein F-actin of LM8 cells. Conclusions The mechanism of zoledronic acid to inhibit vasculogenic mimicry formation of osteosarcoma cells line LM8 may be associated with destruction of ultrastructure in LM8 cells.
