原发性肝癌患者肝组织内HCV RNA核心NS3、NS5区编码抗原的检测

DETECTION OF ANTIGENS RELATED TO HEPATITIS C VIRUS RNA ENCODING CORE.NS3 AND NS5 REGIONS IN THE LIVERS OF PATIENTS WITH HEPATOCELLULAR CARCINOMA

目的研究原发性肝癌(HCC)患者肝组织中 HCV 相关抗原的表达,揭示 HCV 在 HCC 发生中的可能作用。方法应用免疫组化 SP 法定位46例 HCC 组织及38例癌旁组织中 HCV 核心、NS_3、NS_5抗原和 HBsAg。结果癌组织中各抗原的检出率分别为21.7%、21.7%、15.2%和19.6%,癌旁组织中的检出率分别为36.8%、34.2%、31.6%和78.9%。阳性染色细胞呈弥漫、灶状和散在分布.各抗原主要位于肝细胞和肝癌细胞的胞浆内,一些 NS_3、NS_5抗原可位于肝细胞的细胞膜上。阳性染色信号呈细颗粒状,少量的核心和 NS_5抗原可呈均质状.癌旁组织核心抗原阳性表达区可见淋巴细胞聚集,未发现 NS_3和 NS_5抗原表达与病理改变之间的联系。结论 HCV 可能是 HCC 的另一重要致病因素。

Objective By detecting the expression of HCV antigens and HBsAg in the livers of patients with hepatocellular carcinoma(HCC)to elucidate the possible role of HCV in the development of HCC.Method Expression of HCV core、NS_3、 NS_5 antigens and HBsAg in cancerous tissues in 46 patients with HCC and peri-cancerous tissues in 38 patients with HCC were studied by using streptavidin peroxdase conjugataed method(SPmethod).Results The results showed that the detection rates of core.NS_3、NS_5 antigens and HBsAg were 21.7%, 21.7%, 15.2% and 19.6% in HCC group,36.8%, 34.2%,31.6% and 78.9% in peri-cancerous group respectively.The cells with positive staining distributed diffusely,focally and sporadically.The antigens were mainly situated in the cytoplasm of hepatocytes and HCC cells,and some of NS_3、NS_5 antigen at the membrane of hepatocytes.The positive immunoreactive feature appeared in fine granular in appearance,but a small amount of core antigen and NS_5 antigen in feature of homogenous distribution.There have a cluster of lymphocytes accumulation in the area of core antigen expression in peri-cancerous tissues; No associations were found between NS_3 and NS_5 antigens'expression and pathological changes.Conclusion HCV infection might be another important pathogenic factor of HCC and be associated with the development of HCC in the southwest part of china.
