摘要
目的体外细胞毒实验发现重组人肿瘤坏死因子α(rHTNFα)可直接杀伤体外培养的人肝癌细胞株 H_(7402),其抑瘤效应与浓度正相关(r=0.929,P值<0.05);变异性白细胞介素2(IL-2)对 H_(7402)无直接杀伤作用,但可增强 rHTNFα的抑瘤效应,此协同作用呈剂量依赖性:2 500μ/ml 的变异性 IL-2即可明显增强 rHTNFα(1000μ/ml)对肝癌细胞的杀伤力,再加大 IL-2用量,此作用不增加(P值<0.01),rHTNFα升至5万μ/ml 时,只有增加 IL-2用量至2万μ/ml,才出现协同抑制效应。
Objective To obverse effect of TNFα and mutant IL-2 on the hepatic carcinoma cell line in vitro.Methods The hepatic carcinoma cell line(H_(7402))was used as a cell culture system in-vitro.Results The results showed that the inhibition effect on H_(7402) became stronger with the increase of the concentration of rHTNFα(r=0.909,P<0.05) .Mutant IL-2 had no direct inhibition effect,but could enhance the effect of rHTNFα on inhibiting H_(7402) cell line. The concentation of both rHTNFα and mutant IL-2 affected this enhancement:2500U/ml of mutant IL-2 can increase cytotoxity of rHTNFα(1000U/ml)effectively to H_(7402) cell line.When rHTNFα reached 50000U/ml,the enhance effect was appeared only when the concentration of mutant IL-2 was at 20000U/ml.Conclusion result indicated that a rHTNFα combinded with a mutatant IL-2 in low conantration can yield good inhibiting effect on hpatic carcinoma.
