兔中性粒细胞防御素的抗出血热病毒研究 被引量:1

Direct Inactivation of Epidemic Hemorrhagic Fever Virus by Defensins from Rabbit Neutrophils

摘要 从新西兰白兔的中性粒细胞胞浆颗粒中提取兔防御素,测定其体外抑制流行性出血热病毒(EHFV)的活性实验,结果表明:免防御素对EHFV感染Vero细胞无阻断作用,而对EHFV有直接杀灭作用,20μg/ml时其抗病毒指数为196。同时,兔防御素对EHFV在Vero中的增殖有抑制作用,40μg/ml时其抗病毒指数为208。 Defensins are the major component of antibiotic peptides of macrophages and granulocytes.They play an important role in oxygenindependent microbicidal mechanisms.Defensins exert in vitro microbicidal activity against many bacteria、fungi、leptospires and treponema.In addition, they directly inactivate certain enveloped viruses. It was purified for rabbit defensins (NP, mixture of NP 1、NP 2、NP 3a 、NP 3b 、NP 4、NP 5) from rabbit peritoneal exudate neutrophils and tested for their ability to inactivate epidemic hemorrhagic fever virus (EHFV 76118 strain) in vitro.EHFV was susceptible to direct neutralization by NP.When 100TCID 50 /ml of EHFV was incubated with 20 μg/ml of NP at 4 ℃ for 2 hour, more than 50% of viruses were killed, anti virus index (anti virus index=LogTCID 50 of virus control group/LogTCID 50 of NP group) was 1 96.Similarly, when Vero cells were cultured in Eagle medium containing 40 μg/ml of NP, titer of EHFV that grew in Vero was reduced, anti virus index was 2 08.
