摘要
研究了基于树枝状化合物(PAMAM)界面的PCR,将引物通过静电作用连接到PAM-AM分子的表面,采用质粒pB luescript SK作为模板,进行PCR,并用琼脂糖凝胶电泳和透射电子显微镜对PCR产物进行检测,分别研究了退火温度、引物浓度对PCR的影响.实验结果表明:连接在PAMAM分子表面引物的PCR可以成功地进行,并且在一定温度范围里,PCR几乎不受退火温度的影响;通过PCR制备了PAMAM-DNA的聚合体,这种聚合物在高温时能分散,低温时能复合.基于PAMAM分子表面的PCR将均相体系中的PCR技术扩展到非均相体系中,大大扩展了PCR技术的应用范围,提供了一种制备纳米材料的新方法.
A polymerase chain reaction (PCR) procedure based on Polyamidoamine Dendrimers (PAMAM) was systemically studied. Such PCR can proceed successfully under optimized condition, with both the two primers bound to the PAMAM surface, respectively. In certain temperature scope, PCR is insensitive to annealing temperature. The nanostructured aggregates prepared via PCR can disperse and congregate reversibly at desired temperatures. The present PCR procedure can extend its use from liquid homogeneous system to nanoparticle surface.
