Role of Autophagy in the Pathogenesis of Alzheimer’s Disease

摘要 自噬是真核生物中的一种维持细胞基本功能的生命现象。其基本分子机制及调控信号传导通路非常复杂。近年来的研究显示自噬对于致病性β-淀粉样蛋白(Aβ)和细胞骨架相关tau蛋白的生成和代谢都有密切的关系,且其对神经元的死亡也有特殊的作用。随着自噬的生理机制不断被阐明,其在阿尔茨海默病中的作用将得到更广泛的重视。 Autophagy is a basic life phenomenon existed in eukaryote which serves to maintain the homestasis m cells. Although the main physiological and signal transductional pathway was a partly covered during these years, its true activity remained a secret. However, recent findings suggest that autophagy plays an important role in the formation and degradation of β-amyloid and tan, the two important proteins which are closely related to the pathogenesis of Alzheimer's disease. Moreover, autophagy also exists a critical and special influence on the neuron death. It is strongly believed that the role of autophagy in the pathogenesis of Alzheimer's disease will be disclosed.
