SIRT1对内分泌与代谢的调节

摘要 沉默信息调节因子2(SIR2)发现于酵母细胞中的,其参与酵母交配型基因沉默、端粒区基因沉默和重组,维持基因的稳定性,并与物质代谢和长寿有着很大的关系。后来发现SIR2在转录调节过程中具有很关键的催化作用即组蛋白去乙酰化酶活性。进一步研究发现SIR2特别是哺乳动物的同源基因SIRT1在调节内分泌信号方面起着重要的作用。本文主要对SIR2在insulin/IGF-1通路、肝脏、白色脂肪组织、特别是在胰岛β细胞内分泌与代谢的调节作用做一综述。 Silent information regulator 2(SIR2)was discovered in yeast cell. It participates in mating type gene silencing, telomere gene silencing and rDNA recombination. It retains genic stability and takes a important role in substance metabolism and longevity. Latter, histone deacetylation, the important catalysis of SIR2, was discovered in transcription. SIRT1, the homologous gene of SIR2 in mammalian,plays a significant part in regulation of endocrine siguahng. This article reviewed the effects of SIR2 in endocrine and metabolism of insulin/IGF- 1 pathway, liver, white adipose tissue, particularly, beta cell of islet.
